Protective Effect of Buyang Huanwu Decoction on Cerebral Ischemia/Reperfusion Injury by Inhibiting Autophagy Based on AMPK/UlK1 Signaling Pathway

The aim of the paper is to explore whether AMPK/UlK1 signaling pathway involved in regulating autophagy and alleviating cerebral ischemia/reperfusion (I/R) injury. On this basis, study regulatory effect Buyang Huanwu Decoction (BHD) on pathway, aiming clarify molecular mechanism BHD I/R injury protective containing serum (BHDS) OGD/R neurons. research group adopted following methods: Neurons cultured for 7 days vitro were randomly divided into normal control group, model (OGD/R), AMPK activator (AICAR) (3 mmol/L), medicated (3%), + (BHDS AICAR) group. protein expressions p-AMPK, AMPK, p-UlK1, UlK1, Beclin-1 LC3B neurons detected by Western blot, fluorescence intensity apoptosis rate neuron autophagosome CYTO-ID staining flow cytometry, morphological changes immunofluorescence double labeling method, observed inverted microscope, survival was CCK-8 method. Neuron optimal concentration BHDS. subjected hypoxia glucose deprivation 1 h, 2 3 4 h reoxygenation 24 h. release l DH determine successfully established. And results show that BHDS significantly increased, decreased. Among them, 3% intervention at same time best condition. In levels p-AMPK/AMPK, p-UlK1/UlK1, beclin-1, intensity, transformation from LC3B-Ⅰ LC3B-Ⅱ So we can draw a conclusion inhibit alleviate through pathway.
